GENERAL SUMMARY/ OVERVIEW STATEMENT:
The Musolino Laboratory within the Center for Genomic Medicine at Massachusetts General Hospital and Harvard Medical School, a translational neuroscience laboratory focusing on developing gene therapy for inherited cerebrovascular disorders that lead to early onset of stroke and white matter injury (leukodystrophy) seeks an exceptional post-doctoral fellow to lead bench work involving in vitro cell culture and animal models of these neurogenetic diseases. While the candidate will receive support from current study staff, as well as guidance from the Principal Investigator, ample experience and independence to establish the use of molecular biology, iPSC cell complex assays and animal behavioral and surgical techniques is required. Strong communication and people skills are required as well as a desire to work in a fast-paced, highly collaborative, translational research group working at the interface of neuroscience, stem cell biology, and novel therapeutic development. This position is ideal for a candidate looking to pursue an independent academic career or translational research in biotech.
PRINCIPAL DUTIES AND RESPONSIBILITIES:
The post-doc will be primarily responsible for the design, supervision and implementation of the laboratory’s research studies. Responsibilities include:
- Prepares basic solutions and performs base-level procedures as assigned (i.e. pipetting, cell and tissue culture, etc.).
- Maintains laboratory notebook, conducts data analysis, and interpretation of results.
- Handling animals (mice), this includes setting up breeding pairs, performing tail clips and genotyping, behavioral testing and sacrificing for histological examination. Supervise two laboratory research assistants on animal and other laboratory task.
- Conduct experiments utilizing various biochemical and molecular biology techniques:
- Cloning: must be able to independently perform genotyping, design and testing of primers and planning and executing point mutagenesis experiments.
- Gene editing: must be familiar and able to design primers and plasmids under supervision for gene editing/deletion using CRISPR Cas and base editor systems.
- Western blots: independently design and trouble shoot quantitative Western blot experiments.
- Quantitative PCR: must be able to design and test primers as well as plan and execute experiments to quantify mRNA expression levels.
- Enzyme assays: Independently establish the use of new biochemical assays to measure metabolite levels from cultured cells or animal/patient samples.
- Must have experience with microscopy and live cell imaging techniques.
- Experience in mammalian cell culture, especially iPSC-derived models, astrocytes, pericytes/smooth muscle cells, fibroblasts, endothelial cells and/or transformed cell lines is necessary. Will also be required to independently establish the culture of primary cells lines new to the lab.
- Must have experience with surgical techniques in rodents(mice).
- Collection, fixation, sectioning and staining of histological specimens
- Performs independent literature searches and contribute new ideas to the project. Contributions to intellectual property is recognized by direct participation on patent filing.
- Must be able to independently formulate hypothesis and demonstrate a rational approach to experimentation in weekly lab meetings.
- Organizes and summarizes acquired data, using scientific and statistical techniques.
- Organizes and/or drafts material for the preparation of research papers, manuscripts, or other documents for publication and/or presentation.
- Participates in the design of experiments or field work.
- May develop modifications of existing assay techniques, experimental protocols, or new procedures.
- Clearly communicate research results with collaborating labs and present findings at scientific conferences.
- Orients and trains new Research Technicians and Fellows
- During the three years, the post-doc is expected to lead at least one independent project and apply for post-doctoral, career development fellowships/awards as early investigators.
- Must have experience in managing projects.
- Must have experience supervising students and research assistants.
- Must be highly organized, detail-oriented, and have a strong motivation to learn.
- Must have excellent interpersonal, speaking, and writing abilities.
- Must be able to perform multiple tasks independently.
- Must be highly motivated and persistent.
- Ability to troubleshoot technical problems.
- Understanding of transgenic mouse generation.
- Ability to work independently and as a team member.
- Synthesizes and interprets results.
- PhD in life sciences or bioinformatics is required.
- 4+ years research experience.
- 2+ first authored peer review research articles.
The post-doc will spend 5 days per week in Dr. Musolino’s laboratory in the Simches Research Building at Charles River Plaza. Work is divided between the bench on the 5th floor and the animal facility in the Their Building (5 minute walk). Travel to national and international conferences where data is being presented is expected and expenses fully covered. Various core facilities are in close proximity. Occasional visits to MGH-Main Campus and Ragon Institute of MGH, MIT and Harvard maybe required for collaborative studies and utilization of different research cores. The post-doc will work under the direct supervision of Dr. Musolino (PI) and in addition receive training in the laboratories of Drs. Steven Harggarty (expert in iPSC derived-systems in neurodegeneration), Eng Lo (expert in neurovascular unit biology) and Dr. Benjamin Kleinstiver (expert in gene editing) faculty members in the Centers for Genomic Medicine and Neurology Departments.
